We recently developed a new radiolabeled RGD-peptide for detection of αvβ3-integrin using positron emission tomography (PET).HypothesisThe new PET-tracer,Ga-NODAGA-E[c(RGDyK)]2,can detect myocardial angiogenesis after induction of myocardial infarction in vivo.MethodsFour Sprague-Dawley rats had myocardial infarction induced with surgical ligation of the left anterior descendent artery (LAD). Eight and 36 days post myocardial infarction, the animals were injected with 36 [10-81] MBq of Ga-NODAGA-E[c(RGDyK)]2 and scanned using a Siemens preclinical PET/CT Inveon MM system. Furthermore, 9 days after operation, the animals were injected with 100 MBq Tc-Sestamibi and underwent myocardial perfusion imaging using Mediso Nanoscan to verify location and extent of infarction. RGD uptake and perfusion were quantified using INVIA 4DM software v2017.ResultsAll rats had large anteroseptal myocardial infarctions (mean summed rest score 24±13). Uptake of RGD was significantly higher in the infarct compared to non-infarct area one week after myocardial infarction (SUVmean 6.1 vs 4.4, p=0.007). One-month post MI the difference declined (SUVmean 5.0 vs 4.2, p=0.09) (Figure). When analyzed using the 17-segment model there was a significant negative correlation between segments with hypoperfusion by SPECT and angiogenesis by RGD-PET (r= -0.253, p=0.038), this translated into significantly higher uptake of RGD in segments with myocardial infarction compared to segments with normal perfusion (p=0.01)ConclusionsGa-NODAGA-E[c(RGDyK)]2PET can detect αvβ3-integrin expression and thus myocardial angiogenesis following myocardial infarction.
