Summary:Poly(2‐oxazoline)s are water‐soluble synthetic biocompatible polymers. They are typically prepared by living cationic ring‐open polymerization. Depending on the chemical nature of the cationic initiators, two distinct polymerization mechanisms have been identified: ionic and covalent propagation. A wide variety of structural variability can be achieved by the monomer selection influencing the hydrophilicity/hydrophobicity and the living nature of the polymerization allows for the formation of block and gradient copolymers. Poly(2‐oxazoline)s are pseudopeptides and the similarity to proteins is the basis for their exceptional biocompatibility. Poly(2‐oxazoline)s started to replace poly(ethylene glycol)s in their typical fields of application, such as drug delivery, hydrogels, and surface modification.
