Summary:The synthesis of 4-(p-biphenyl)-1-(2H)phthalazinone (1) is described. Acylation of 1 using acetic acid/acetic anhydride mixture gave 2-N-acetylphthalazinone 2a and 1-acetoxyphthalazine 2a'. Condensation of ea with aromatic aldehydes gave 2N-cinnamoyl-1-(2H) phthalazinone 3a-d. Reaction of 1 with nitrogen nucleophiles such as semicarbazide, thiosemicarbazide and hydroxylamine hydrochloride yielded 4 and 5. The lactim form of 1 is also investigated through its reaction with ethyl chloroacetate. The reaction of the ester derivative 7 with hydrazines, amines, aromatic aldehydes (under Stobbe conditions) has been described. The Mannich reaction of 1 was also investigated. Structural assignments of the new products were based on elemental analysis and spectral data.
