Summary: | The present invention relates to a system for the regulation of gene expression. The invention provides improved nucleic acid constructs capable of tightly regulating the expression of a coding sequence of interest. Tight regulatory control is desirable where the nucleic acid to be expressed, or the level of expression, is toxic to the cellular or host environment in which expression occurs. The invention also relates to the application of the expression system in cells used to package viral vectors and provides methods of preparing the necessary nucleic acid constructs as well as their use in the control of recombinant viral gene expression. In one aspect, the invention relates to the regulation of gene expression in a stably transfected cell.
The present invention provides nucleic acid constructs, expression systems, and methods relating to the regulation of gene expression. The invention may be applied to regulate the expression of any coding sequence of interest, including those coding for viral components necessary for the packaging of viral particles. |
