Summary:Traditional high-performance liquid chromatography (HPLC) method development simply required careful definition of column, mobile phase and detector conditions. However, for ultrahigh-pressure liquid chromatography (UHPLC), which offers very high column efficiency and narrow sample elution bands, the UHPLC instrument components must be carefully matched in performance with columns to achieve desired resolution. If UHPLC underperforms, it can be difficult to determine whether the instrument or column is the problem. This article will describe the design of modern UHPLC instruments and offer simple procedures to identify the causes of poor column and method performance.
