Summary:Objective To study the effect of the EGFRv III siRNA on the expression of EGFRv III and the apoptosis and cell growth in the U87-EGFRv III cell line. Methods The EGFRvl siRNA was synthesized by the chemical method. The gene expression of EGFRv III was detected by RT-PCR and the protein expression was detected by Western blot and immunofluorescence. MTT and Annexin V-FTTC/PI assays were used to detect the inhibitory effect on cell proliferation and the apoptosis effect Results The results of RT-PCR and Western blot showed that the gene and protein expression of EGFRvITJ was decreased significantly by EGFRv III siRNA, which could be reduced (75.7 plus or minus 3.1)% after transfected with siRNA for 48 h. The results of the MTT and Annexin V-FITC/PI assays showed that siRNA could suppress the cell growth of glioma cell line and induce apoptosis of the glioma cell. Conclusion EGFRvK siRNA could significantly inhibit EGFRv III gene, protein expression and the proliferation, as well as induced apoptosis of U87-EGFR
