Summary:One of the main objectives of proteomics is to identify and quantify proteins in a tissue or a cell. Tandem mass spectrum has been proved to be a powerful tool to achieve this goal. This paper presents a review on the protein identification approaches based on tandem mass spectrometry, including database searching methods, De Novo methods, PST (peptide sequence tag) methods, and spectra library methods. It also introduces the research on peptide fragmentation principle, amino acids neutral losses, and peptide identification. Finally, the viewpoints on the development of mass spectrum techniques are described.
