Summary:Background and aims. Poly ADP ribosylation (PAR) has been correlated with neurotrophin and neuroprotec-tive peptide activity (1). It was also shown that brain derived neurotrophic factor and neurotrophin3 play a role in memory consolidation (2). Our studies showed cognitive enhancement by neuroprotective peptide (NAP) treatment (3). Furthermore, previous experiments reveled that PAR inhibition prevent memory consolidation in the invertebrate Aplysia (4). Our current research is aimed at PAR inhibition (PJ-34) in rodents, questioning whether PAR inhibition impairs memory consolidation processes. Methods. Mice were subjected to intracerbrovascular injection proceeding (45 min later) an acquisition trial session of either an object recognition task or a passive avoidance paradigm. Results. Control mice exhibited significantly higher memory retrieval scoring than mice injected with a PAR inhibitor, when tested in long-term memory paradigms. Conclusion. PAR inhibition impairs memory consolidation in mice. Since our previous work showed that PAR is mediated by neurotrophins and neuroprotective peptides, it is highly probable that neurotrophin-depen-dent memory consolidation is also mediated via PAR.
