Summary:This laboratory has, for some time, been involved in the study of antibody combining sites, particularly those with specificity for carbohydrate determinants. In spite of significant progress through biochemical and molecular biology studies, a fundamental understanding of individual amino acids, or clusters of amino acids, necessary for antigen binding has yet to be achieved. To approach this problem, we are in the process of constructing site directed mutants with alterations at positions predicted to be involved in antigen binding. Herein, we describe the isolation and characterization of a V sub(H) construct from a prototypic galactan binding myeloma, X-24, suitable for such studies.
