Summary:A single dimeric form of malate dehydrogenase is found in Escherichia coli whereas eucaryotic cells contain cytoplasmic and mitochondrial forms of this enzyme. The procaryotic mdh gene was isolated using an oligonucleotide probe synthesized on the basis of partial amino acid sequence information. Restriction fragments within the isolated gene were subcloned into M13 vectors for nucleotide sequencing on both strands using the dideoxy chain termination method. The nucleotide sequence of the mdh gene and the derived amino acid sequence for malate dehydrogenase are shown.
