Summary:Objective: To observe the possibility that the proteomics of the tumor interstitial perfusing fluid can be analyzed as the tumor microenvironment proteomics. Methods: Clean fresh tumor tissue biopsies were incubated in phosphate buffered saline (PBS). The tumor microenvironment proteins were extracted in perfusing fluid and then analyzed by proteomics technique. Results: The proteins in tumor interstitial perfusing fluid were analyzed by two-dimensional gel electrophoresis (2DE), Matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF-MS) and bioinformatics; one of the proteins was identified as secreted protein Profilin-1. Conclusion: The tumor interstitial perfusing fluid proteomics can be analyzed as the tumor microenvironment proteomics. 目的 探讨肿瘤间质灌注液蛋白质组作为分析肿瘤组织中细胞微环境蛋白质组的可能性。方法 应用PBS液孵育新鲜肿瘤组织,提取肿瘤间质灌注液中的总蛋白,并应用蛋白质组学技术对其进行分析研究。结果 应用双向电泳、质谱和生物信息技术成功分离并鉴定出肿瘤间质灌注液中的蛋白成分;在肿瘤间质灌注液中发现肿瘤分泌蛋白Profilin-1。结论 肿瘤间质灌注液蛋白质组学研究为研究肿瘤组织中细胞微环境蛋白质组提供了新的研究方案。
