Summary: | 2.2 Reagents Herba Lycopi (collected in Youjiang District of Baise City) ; cellulase (activity 40000 U/g) (Tianjin Lihua Enzyme Preparation Co. , Ltd. ); glucose (Guangzhou Aixin Scientific Instrument Co. , Ltd. ) ; DPPH (Sigma Company) ; other reagents were of analytical grade. 3 Methods and results 3.1 Drawing of standard curve 50 mg of standard glucose sample dried (105 °G ) to constant weight was accurately weighed, and the distilled water was used for preparation of 100 ml standard glucose solution. 0, 2, 4, 6, 8, 10 ml of standard glucose solutions were suctioned, respectively, and the volume was set to 50 ml, to get the dilutions of different concentrations. According to the test results, with the extraction rate of polysaccharides as test indicator, the Lg(34) orthogonal experiment was conducted. 3.4 Detection of extraction rate of Herba Lycopi polysaccharides In this study, we used sulfuric acid-phenol method^3' to determine the polysaccharides. 0.5 ml of test solution was placed in 10 ml test tube, and mixed with 1.0 ml of 5% phenol solution and 5.0 ml of concentrated sulfuric acid. The analysis of variance showed that enzyme dosage and enzymolysis temperature had obvious significance in the extraction of Herba Lycopi polysaccharides.
