Summary:By utilizing a rat model of acute lung injury, we demonstrated that inflammatory response induced by IL-1ß instillation is accompanied by upregulation of p38a signaling and is counteracted by intratracheal administration of the novel potent and selective p38a inhibitor, CHF-6297 (CHI-91040). Intratracheal recombinant rat IL-1ß dose dependently increased neutrophil numbers and IL-6 levels in bronchoalveolar lavage (BAL), a phenomenon accompanied by p38 MAPK signaling activation as detected by phosphorylation of HSP27 in lung homogenates.
