Summary: | We have demonstrated that oxidation of nucleotides in nucleotide pools, as well as direct oxidation of DNA, causes mutagenesis and cell death, the former resulting in carcinogenesis while the latter in neurodegeneration. Under environmental stress, various damaged nucleotides are generated in nucleotide pools by oxidation, deamination and halogenation. We purified proteins that specifically bind such damaged nucleotides from cultured human or mouse cells, and identified enzymes involved in hydrolysis of various damaged nucleotides. In this workshop, we will present mechanisms for the maintenance of nucleotide pool homeostasis, and molecular pathologies associated with its disruption. |
