Summary: | Effects of CuDIPS, a chemical having SOD like activity, on the reduced status of GSH, the major nonprotein thiol in tissues, were investigated using rat freshly isolated hepatocytes. CuDIPS below 100 uM did not have any significant effects both on the GSH content and viability of the hepatocytes but at 150-250 mM it decreased both of them dose dependently after 1hr of incubation. The decrease in cellular GSH did not seem to be followed by an increase in the GSSG in the cell suspension. The addition of deferoxamine, an iron chelator, with CuDIPS to the hepatocyte suspension protected depletion in GSH content and loss of cell viability by CuDIPS. CuCl_2 at 200uM decreased cellular GSH content only slightly. It appears that CuDIPS penetrated through the cell membrane and acted to decrease the GSH level by forming a copper-glutathione complex. |
