Summary:The anti-hypercholesteremic properties of KYKC-170, a choroman derivative, were investigated from the viewpoint of cholesterol absorption and excretion in mice, and cholesterol metabolism in rats with the following results. 1)KYKC-170 significantly reduced the serum cholesterol(CH) level in mice treated high fat emulsion. 2)KYKC-170 significantly reduced the CH level, increased the HDL-CH level and decreased the (CH -HDL-CH)/HDL-CH ratio designated as the "atherogenic index" in mice fed high CH diet. 3)KYKC-170 had ,no effect on the ill situ uptake in the rat small intestine of ^^14 C-CH given the lumen. 4)The oral pre-treatment of the animals with KYKC-170 supressed the in vitro incorporation of ^^14 C-acetate into CH, In liver slices from normal and Triton-treated hypercholesteremic rats 5)The kinetics of serum levels of ^^14 C-CH given orally and of ^^3 H-CH given intravenously revealed that KYKC-170 when orally administered, had no effect on the absorption of ^^14 C-CH from the digestive tract nor the excretion of serum ^^3 H-CH.
