Summary:With the aid of in situ hybridization the localization of a cluster of genes in the early ecdysone-inducible puff 2B region of Drosophila melanogaster was determined in eight species of Drosophila. The site of localization of genes ecs, dor and swi within this cluster is the puff at the telomere end of the X-chromosome in D. funebris, D. virilis, D. hydei, D. repleta, D. mercatorum, and D. paranaensis. In D. kanekoi the site of localization is the puff in the distal region, and the site of localization in D. pseudoobscura is the proximal region of the X-chromosome. It is proposed that this cluster has a conservative organization in the above species. The dispersed distribution of certain DNA sequences from the given region in D. hydei chromosomes was observed
