Summary:Leaf explants of in vitro grown tobacco plants were transformed with two different binary vectors using Agrobacterium-mediated transformation. These two binary vectors pUN1 and pJL03 differed in bacterial copy number. For each vector, the transformation was performed with two different concentration of agrobacteria. No obvious differences in transformation efficiency and speed were noted. GUS activity was determined as a measure for the level of GUS gene expression by quantitative fluorometric GUS assays. The measurements showed that the vector type and bacterial density are less influential than anticipated.
