Summary:In this study, investigations were carried out to isolate, identify and characterize infectious laryngobracheitis virus (ILTV) isolates from commercial layer chicken farm located at Lipa City, Batangas [Philippines]. The virus was isolated using chorio allantoic membrane (CAM). A total of five sample (IV, VI-C28, VI-C29, VI-C30, and VII) were produced typical plaque lessions in CAM at second level passages such as yellowish plaques with opaque edges. The plaque lessions were also distributed throughout the CAM. The positive infected ILTV on CAMs were confirmed by Agar Gel immuno-diffusion (AGID) test, using antisera standard reference against ILT (Anti-NS175). The AGID test with specific antisera revealed that a-five isolates from Batangas were reacted with the specific antisera reference and produced the line identity reaction. Furthermore, a-five isolates were then characterized by immuno blotting on 7.5% of acrylamide. These results showed that isolates VI-C28, VI-2a, VI-30 and VII have four major viral protein of 205, 160, 85 and 60 kDa, respectively and each viral proteins bind to each antiviral antigens. This findings indicated that isolates VI-C28, VI-C29, VI-30 and VII have homologous viral protein and each other were identical epitope. Furthermore, the vaccine strain (BAL-ILT) produced 3 major viral protein of 160, 85 and 60 kDa, and all of them reacted to antiviral protein of isolates VI-C28, VI-29, VI-29, VI-30 and VII. However, the isolate IV only has two major viral protein (205 and 85 kDa) and these viral proteins were homologous with viral protein of 205 and 85 kDa of isolates VI-C28, VI-29, VI-30 and VII. ILT structural proteins of 205, 160, 85 and 60 kDa are major ILTV immunogens. Antibodies directed against major immunogens probably play important roles in protective immunity
