Summary: | Asiatic bitter yam (D. hispida) is one of the chief food of tropical Asia during times of food scarcity. The tuber is poisonous because of a high content of alkaloid dioscorine, therefore it can also be used for natural pesticide. D. hispida is mostly gathered from the wild, and there are hardly any production statistic. In vitro maintenance is one alternative to keep the continuous availability of D. hispida for further research and usage. Research was conducted at tissue culture laboratory, Research Institute for Food Crop Biotechnology in the fiscal year 1998/1999. Two trials were done as follows: 1. combination of basic medium (Anderson and WPM) with IAA (0.1 mg/l) and Thidiazuron (0.1 mg/l), and 2. combination of basic medium (Anderson and WPM) with 2 levels of BA concentration (0.5 and 1.0 mg/l). The result showed that Anderson basic medium was better than WPM for D. hispida propagation purpose. The effect of IAA enrichment were not significantly different from Thidiazuron enrichment on shoot number per plant, plant height and number of node per plant. Addition of BA from 0.5 mg/l up to 1.0 mg/l on Anderson medium increased shoot number per plant, plant height, however reduced node number per plant
Gadung merupakan salah satu ubi-ubian yang dapat digunakan sebagai makanan alternatif pada masa-masa sulit. Selain itu tanaman ini dapat digunakan sebagai pestisida alami. Karena itu koleksi tanaman gadung secara kultur in vitro merupakan salah satu alternatif untuk melestarikan tersedianya tanaman agar dapat dibudidayakan. Penelitian dilakukan di Laboratorium Kultur Jaringan Kelti Reproduksi dan Pertumbuhan Balitbio Bogor pada tahun anggaran 1998/1999. Perlakuan terdiri dari 1. kombinasi antara media dasar (Anderson dan WPM) dengan IAA (0,1 mg/l) dan Thidiazuron (0,1 mg/l) dan 2. kombinasi antara media dasar (Anderson dan WPM) dan konsentrasi BA (0,5 dan 1,0 mg/l). Hasil penelitian menunjukkan bahwa media dasar Anderson lebih baik dari media WPM dalam perbanyakan tanaman gadung. Pengaruh penambahan IAA tidak berbeda dengan penambahan Thidiazuron terhadap jumlah tunas/tanaman, tinggi biakan dan jumlah buku/tanaman.
