Summary:A double antibody sandwich-ELISA method for the titration of Aujeszky's virus was elaborated, and on this basis a method suitable to determine the antibodies was developed. Cytopathic titres of virus suspensions produced in various cell cultures were compared with the total antigen concentration measured with ELISA. The cytopathic titre and the total antigen concentration measured with ELISA showed a good linear correlation in each cell culture. For the specific determination of the anti-Ay antibody level the inhibitory ELISA is by all means more suitable than the indirect method. The sandwich-ELISA system does not detect the contaminations of the virus suspension, and the measured reduction of extinction is exclusively caused by the reaction of the antiviral antibody with the virus. Thus, the purified antigen is not required.
