Summary: | Five peptides were designed according to the genetic/amino acidic sequence profiles of circulating small ruminant lentiviruses (SRLV) from sheep of North-Central Spain. The ELISAs produced with these peptides and two commercial ELISAs used as controls were tested against a panel of 539 sera, involving animals from controlled flocks as well as SRLV-induced arthritis (genotype B2) and encephalitis (genotype A) outbreaks, and animals from flocks infected with different SRLV types. The results obtained indicate that a proportion of animals seronegative to commercial tests is seropositive to one or more synthetic-peptide ELISAs. This proportion varied between flocks and between peptides. None of the commercial ELISA tests or the peptides ELISAs was able to detect all the infected animals in the studied population. One peptide ELISA (with consensus A sequence of SU V5 region of circulating SRLV) detected seropositives in most animals in the nervous outbreak (sensitivity and specificity above 90% and 84%, respectively), all the visna affected sheep tested from this outbreak and only 9% of the arthritic animals from the arthritic outbreak. Two other peptide ELISAs had different reactivity patterns in caprine arthritis encephalitis type (CAEV) and Visna/ Maedi (VMV) type infections which may be useful to genetically classify flock infection. A choice of the diagnostic test, based on the genetic/antigenic type of local circulating SRLV for the detection of seropositives, as well as the application of additional tests to detect infection amongst seronegatives, are essential to reach the SRLV-free status of the flock. |
