Summary: | The active alpha-amylase and Cx-cellulase of Aspergillus niger mutants were exposed to second mutagenic treatments by 5-bromouracil (5-BU) and 2-amino purine (2-AP) with different concentrations to improve these enzyme productions. The qualitative and quantitative measurement of enzyme activity of 30 mutants obtained after 5-BU treatment and seven mutants obtained after 2-AP treatment revealed a considerable improvement in enzyme production. The effects of using different media, carbon and nitrogen sources in the minimal media have been tested after performing the second mutagenesis. The use of dextrin yeast extract medium (DYE) enhanced the alpha-amylase production to 2.5 mg/ml filtrate and carboxymethyl-cellulase (CMC) medium activated Cx-cellulase production to 3.55 u mol/min.
