Summary: | A total of 20 Slovak beers produced from the 2003 barley harvest by different companies were analysed for fumonisin B1 and B2 contamination. A method based on immunoaffinity column extraction and clean-up, coupled with HPLC analysis with fluorescence detection after derivatisation with a mixture of o-phthaldialdehyde and 2-mercaptoethanol, was used. The detection limit for both fumonisins was 1 microg/mL, the recovery was 93% for fumonisin B1 and 78% for fumonisin B2. Phosphate buffer usually applied resulted in a poor separation of derivatised fumonisins. Peak splitting depended on the pH of the eluent. The pH value of 2.6 was found suitable for the peak splitting elimination. A convenient gradient elution metod was suggested to avoid possible interference in the determination of fumonisin concentrations. Fumonisins concentrations were under the limit of detection in all the beer samples tested. |
