Summary:The effect of nutrient medium on growth of Escherichia coli containing the 13 kbp pBI 121 plasmid and on yield of isolated plasmid DNA were examined. Six nutrient media (LB, SOB, SOC, King B, MPA(g) and YDC) were tested in five repetitions. The highest bacterial biomass after 290 min of cultivation was observed in the media LB (OD sub(550) 0.745), King B (OD sub(550) 0.647) and SOB (OD sub(550) 0.553). The highest yield of plasmid DNA was obtained from E. coli cultivated in MPA(g) medium (305 ng/ml). The yields from King B (150 ng/ml), SOC (113 ng/ml) and SOB (84 ng/ml) were also higher than the yield from LB medium (47 ng/ml). We conclude that the best media to culture E. coli with large plasmids such as pBI 121 are those enriched with meat extract and glucose (MPA(g)) or nutrient media enriched with Mg2+ and PO43- ions (King B).
