Summary:Two serological tests, double antibody sandwich ELISA (DAS-ELISA) and enzyme-amplified ELISA (AMP-ELISA) were used to detect beet mild yellowing virus (BMYV) in aphids (Myzus persicae). AMP-ELISA appeared to be more sensitive and reliable for detection. Absorbance values of AMP-ELISA were approximately twice those with standard ELISA. Reliability of BMYV detection was higher after 48 h of acquisition time than with 24 h acquisition. BMYV was reliably detected in samples prepared from groups of four and more aphids. BMYV was not detected in individual aphids after 24 h of acquisition, but two out of five aphids were found to be BMYV positive after 48 h of acquisition. While AMP-ELISA is a more suitable method for detection of BMYV in aphids, low virus concentrations in aphids make a reliable detection of BMYV in individual aphids impossible.
