Summary: | Attempts were made to culture leaf and root tip explants to obtain protocorm like bodies (plbs) as it is normally produced in shoot tip and axillary bud. Plbs from the leaf tips of Spathoglottis plicata, Vanda roxburghii and Rhynchostylis retusa were successfully produced in modified semisolid VW medium (V(6) A(9)). Callus/plbs from the root tips of V. roxburghii, R. retusa and S. plicata were obtained within 25-35 days in an irregular manner in V(6) A(9). Plbs derived from leaf tip of V. roxburghii resembled flower bouquet. Plantlets differentiated from leaf tip derived protocorm of S. plicata and were successfully transplanted into specially prepared potting media. Protocorms of V. roxburghii differentiated into shoot. Protocorms of R. retusa were subcultured in V(6) A(9) medium to obtain multiple shoots. |
