Summary:The present invention relates to methods and compositions useful in the treatment and prevention of Hepatitis C virus (HCV) infections and the symptoms and diseases associated therewith. In particular the present invention relates to DNA vaccines that encode the HCV Core protein and a polynucleotide sequence that encodes at least one other HCV protein, wherein the vaccine causes expression of the proteins within the same cell and the sequence of the polynucleotide sequence encoding the core protein has been mutated or positioned relative to the polynucleotide sequence encoding the at least one other HCV protein such that the negative effect of expression of the Core protein upon the expression of the said at least one other HCV protein is reduced.
