Summary:Provided is a genetically engineered human pluripotent stem cell line in which an insertion sequence including a nucleotide sequence of a fluorescent reporter gene is inserted following a stop codon of the TUBB3 gene, whereby the TUBB3 gene and the fluorescent reporter gene are co-expressed. Genetic manipulation is made to express the fluorescent reporter gene in response to the expression of the TUBB3 gene, a neural cell marker, so that when the cell line is differentiated into neural cells, the fluorescent signal is observed, thereby allowing for monitoring the differentiation process. L'invention concerne une lignée de cellules souches pluripotentes humaines génétiquement modifiées dans laquelle une séquence d'insertion comprenant une séquence nucléotidique d'un gène rapporteur fluorescent est insérée à la suite d'un codon d'arrêt du gène TUBB3, le gène TUBB3 et le gène rapporteur fluorescent étant co-exprimés. Une manipulation génétique est effectuée pour exprimer le gène rapporteur fluorescent en réponse à l'expression du gène TUBB3, un marqueur de cellule neuronale, de telle sorte que lorsque la lignée cellulaire est différenciée en cellules neurales, le signal fluorescent est observé, ce qui permet le suivi du processus de différenciation. TUBB3 유전자의 정지코돈 뒤에 형광리포터 유전자 염기서열을 포함하는 삽입서열이 삽입되어 TUBB3 유전자 및 형광리포터 유전자가 함께 발현되는 유전자 조작된 인간 전분화능 세포주를 제공한다. 신경세포 마커인 TUBB3 유전자의 발현에 의해 형광리포터 유전자가 발현되도록 유전자 조작을 하여 세포주를 신경세포로 분화시킬 때 형광신호를 관찰함으로써 분화 과정을 모니터링할 수 있다.
