Summary:The present invention relates to a self-priming hairpin-mediated isothermal amplification (SP-HAMP) and, more specifically, to a hairpin probe having a self-priming structure, which is for detecting a target nucleic acid and can be used in the nucleic acid isothermal amplification, and to a method for detecting a target nucleic acid by using same. The SP-HAMP technique according to the present invention is convenient due to no need of a separate primer with a complicated design required in existing LAMP technology, has improved efficiency of detection compared with existing LAMP reactions, and can detect DNA as well as RNA as a target nucleic acid, and therefore the technique according to the present invention can be applied in a wider variety of fields. La présente invention concerne une amplification isotherme médiée par boucle en épingle à cheveux à auto-amorçage (SP-HAMP, de l'anglais «self-priming hairpin-mediated isothermal amplification ») et, plus spécifiquement, une sonde en épingle à cheveux ayant une structure à amorçage automatique qui est destinée à détecter un acide nucléique cible et qui peut être utilisée dans l'amplification isotherme d'acide nucléique, ainsi qu'un procédé de détection d'un acide nucléique cible à l'aide de celui-ci. La technique SP-HAMP selon la présente invention est pratique en raison de l'absence d'une amorce séparée avec une conception compliquée requise dans la technologie LAMP existante, présente une efficacité de détection améliorée par rapport aux réactions LAMP existantes et peut détecter de l'ADN ainsi que de l'ARN en tant qu'acide nucléique cible. Par conséquent, la technique selon la présente invention peut être appliquée dans une variété de domaines plus large . 본 발명은 자가 헤어핀 프라이머 기반 등온 증폭 기술(Self-priming hairpin-mediated isothermal amplification (SP-HAMP))에 관한 것으로, 더욱 자세하게는 핵산 등온증폭에 사용할 수 있는 표적핵산 검출을 위한 자가 프라이머 구조를 가지는 헤어핀 프로브 및 이를 이용한 표적핵산의 검출방법에 관한 것으로, 본 발명에 따른 SP-HAMP 기술은 기존의 LAMP 기술에서 요구되는 복잡한 디자인의 별도의 프라이머가 필요로 하지 않아, 간편하며, 기존의 LAMP 반응보다 향상된 검출 효율을 가지며, 표적핵산으로 DNA뿐만 아니라 RNA 검출도 가능하여, 보다 넓은 분야에 적용이 가능하다.
