Summary: | The present invention provides a DNA sample processing method, a sequencing method and a kit. The so-called DNA sample processing method comprises subjecting the DNA, in a first pre-mixed system, to fragmentation, end repair and dA tailing treatment to obtain a first product. The so-called first pre-mixed system comprises pre-mixed enzymes. The so-called pre-mixed enzymes comprise a DNA fragmentation enzyme, a polynucleotide kinase and a DNA polymerase. On the basis of test experiments and an optimized mixed enzyme system, multiple enzymes are mixed in the same reaction system to realize DNA fragmentation, end repair and dA tailing in one step. The method is simple and quick, saves the operating time of sample processing before loading on a machine, and is beneficial to industrialization.
