Summary:The present invention is concerned with novel variant polypeptides having allene oxide synthase activity, and in particular a polypeptide defined by any one of SEQ ID NO: 1, SEQ ID NO: 2, SEQ ID NO: 3, SEQ ID NO: 4, SEQ ID NO: 5, SEQ ID NO: 6 and SEQ ID NO: 7, as well as antioxidant compositions comprising the same. Surprisingly, the novel AOS enzyme variants retain a similar activity and pH optima relative to known AOS enzymes, despite having non-conservative amino acid substitutions at residues that are predicted from crystal structure information to be important to structure/function activity. The variant AOS enzymes according to the present invention may be made by recombinant or synthetic means for use in biological processes of interest. La présente invention concerne de nouveaux polypeptides variants présentant une activité oxyde d'allène synthase, et en particulier un polypeptide défini par l'une quelconque des SEQ ID NO : 1, SEQ ID NO : 2, SEQ ID NO : 3, SEQ ID NO : 4, SEQ ID NO : 5, SEQ ID NO : 6 et SEQ ID NO : 7, ainsi que des compositions antioxydantes les comprenant. De manière surprenante, les nouveaux variants d'enzyme AOS conservent une activité similaire et un pH optimal par rapport aux enzymes AOS connues, malgré des substitutions d'acides aminés non conservatrices au niveau des résidus qui, selon les informations sur la structure cristalline, sont importantes pour l'activité structure/fonction. Les variants d'enzymes AOS selon la présente invention peuvent être fabriqués par des moyens recombinants ou synthétiques destinés à être utilisés dans des processus biologiques d'intérêt.
