Abstract As demonstrated herein, a preferential and progressive expansion of a subset of hematopoietic cells bearing somatic mutations in one or more HSC cardiometabolic driver genes leads to pro-inflammatory signaling at multiple levels. Accordingly, provided herein are compositions, methods, and assays for modulating a HSC cardiometabolic driver gene mutation-mediated proinflammatory activity. Comme démontré dans la présente description, une expansion préférentielle et progressive d'un sous-ensemble de cellules hématopoïétiques portant des mutations somatiques dans un ou plusieurs gènes moteurs cardiométaboliques des CSH conduit à une signalisation pro-inflammatoire à de multiples niveaux. Par conséquent, la présente invention concerne des compositions, des méthodes et des dosages permettant de moduler une activité pro-inflammatoire médiée par des mutations de gènes moteurs cardiométaboliques des CSH.
