Summary: | The present invention relates to a method for screening a protein-protein interaction inhibitor using a nanopore, the method comprising the steps of (a) forming a protein conjugate of a first protein and a second protein, (b) measuring the intensity of an electrical signal generated while the protein conjugate passes through the nanopore, (c) treating the protein conjugate with a protein-protein interaction inhibitor candidate, and measuring the intensity of the electrical signal generated while the protein conjugate passes through the nanopore, and (d) comparing the intensities of the electrical signals measured in steps (b) and (c) to, if the intensity of the signal increases or decreases, determine being a protein-protein interaction inhibitor. The screening method of the present invention enables a large-scale screening of a protein binding inhibitor even with a picomole of a sample without using a label.
La présente invention concerne un procédé de criblage d'un inhibiteur d'interaction protéine-protéine utilisant un nanopore, le procédé comprenant les étapes de (a) formation d'un conjugué de protéine d'une première protéine et d'une deuxième protéine, (b) mesure de l'intensité d'un signal électrique généré pendant que le conjugué de protéine traverse le nanopore, (c) traitement du conjugué de protéine avec un inhibiteur d'interaction protéine-protéine candidat, et mesure de l'intensité du signal électrique généré pendant que le conjugué de protéine traverse le nanopore, et (d) comparaison des intensités des signaux électriques mesurés dans les étapes (b) et (c) pour, si l'intensité du signal augmente ou diminue, déterminer si celui-ci est un inhibiteur d'interaction protéine-protéine.
