In some aspects of the invention, the first universal tail sequence and the second universal tail sequence are different sequences. La présente invention concerne, selon certains modes de réalisation, un procédé de préparation d'un échantillon en vue de son séquençage comprenant les étapes consistant à recevoir un échantillon et à amplifier au moins un marqueur présent dans l'échantillon. Selon certains modes de réalisation, l'amplification du premier marqueur peut comprendre le mélange de l'échantillon avec un premier oligonucléotide comportant une première séquence de queue universelle et un second oligonucléotide comportant une seconde séquence de queue universelle.
