Summary:The present invention pertains to an insecticide compound derived from a strain of Bacillus thuringiensis and relates to the field of plant pest control, particularly to the control of the boll weevil - Anthonomus grandis. More specifically, the subject matter of the invention relates to a new gene for a new delta-endotoxin designated Cry8Ha and to the cloning and expression of the gene that encodes for the protein Cry8Ha in Escherichia coli. The nucleotide and protein sequence that encodes the new delta-endotoxin, recombinant vectors and host cells are disclosed. Also disclosed are processes and means for the recombinant production and use of the new delta-endotoxin for controlling the boll weevil. Additionally, the invention also provides an optimized synthetic gene for expression in cotton plants. Using the gene described herein, it is possible to transform plants using techniques known by specialists in the field for the expression of the endotoxin active against the boll weevil. La présente invention concerne un nouveau composé insecticide dérivé d'une souche de Bacillus thuringiensis, et trouve une application dans la lutte contre des organismes nuisibles pour les plantes, notamment contre le charançon du coton (Anthonomus grandis). Plus particulièrement, la présente invention a pour objet un nouveau gène pour une nouvelle delta-endotoxine appelée Cry8Ha ainsi que le clonage et l'expression dans Escherichia coli du gène codant pour une protéine Cry8Ha. L'invention concerne également la séquence nucléotidique et protéique codant pour la nouvelle delta-endotoxine, des vecteurs recombinants et des cellules hôtes. Elle porte en outre sur des procédés et des moyens pour la production recombinante et l'utilisation de cette nouvelle delta-endotoxine en vue d'une application dans la lutte contre le charançon du coton. Par ailleurs, l'invention concerne un gène synthétique optimisé pour une expression dans les cotonniers. L'utilisation du gène susmentionné permet de transformer des plantes, à partir des techniques connues par les spécialistes dans ce domaine, en vue de l'expression de l'endotoxine active contre le charançon du coton. A presente invenção pertence a um novo composto inseticida derivado de uma linhagem de Bacíllus Thuringiensis e refere-se ao campo de controle de pragas de plantas, particularmente ao controle do bicudo-do-algodoeiro - Anthonomus grandis. Mais especificamente, o objeto da invenção refere-se a um novo gene para uma nova delta-endotoxina designada Cry8Ha e a clonagem e expressão em Escherichia coli do gene que codifica para a protéina Cry8Ha. São fornecidas a seqüência nucleotídica e protéica codificadora da nova delta-endotoxina, vetores recombinantes e células hospedeiras. São ainda fornecidos processos e meios para a produção recombinante e o uso da nova delta-endotoxina para aplicação no controle do bicudo-do-algodoeiro. Adicionalmente, a invenção também fornece um gene sintético otimizado para expressão em plantas de algodão. Usando o gene aqui descrito, é possível a transformação de plantas, a partir das técnicas conhecidas pelos especialistas na área, para a expressão da endotoxina ativa contra o bicudo-do-algodoeiro.
