Summary: | cDNA that encodes a protein expressed on the surface of a cell or secreted from a cell is selected among from a cDNA library derived from a cancer cell strain utilizing an SST-REX method, a monoclonal antibody against the protein encoded by the selected cDNA is prepared, and the bindability to various types of cancer cell strains and the in vitro and in vivo anti-cancer activity of the monoclonal antibody are examined. As a result, a monoclonal antibody that can bind to PODXL2 protein and has an excellent anti-cancer activity can be found. A region containing an epitope in the antibody can be successfully identified, and the amino acid sequences for variable regions in a light chain and a heavy chain of the antibody can also be successfully determined.
Selon l'invention, de l'ADNc qui code pour une protéine exprimée sur la surface d'une cellule ou secrétée à partir d'une cellule est sélectionné parmi une bibliothèque d'ADNc issue d'une souche de cellules cancéreuses utilisant une méthode SST-REX, un anticorps monoclonal dirigé contre la protéine codée par l'ADNc sélectionné est préparé, et la capacité de liaison à divers types de souches de cellules cancéreuses et l'activité anticancéreuse in vitro et in vivo de l'anticorps monoclonal sont examinées.
