Summary:Disclosed is a method for industrially producing a single form of 3-carboxy-cis,cis-muconic acid and/or 3-carboxymuconolactone from a low-molecular-weight mixture derived from a plant constituent and containing vanillin, vanillic acid, protocatechuic acid and the like through a multi-step enzymatic reaction. Specifically disclosed are: a recombinant plasmid carrying a vanillate demethylase gene (vanAB gene), a benzaldehyde dehydrogenase gene (ligV gene) and a protocatechuate 3,4-dioxygenase gene (pcaHG gene); a transformant having the plasmid introduced therein; and a method for producing 3-carboxy-cis,cis-muconic acid and/or 3-carboxymuconolactone, comprising the step of culturing the transformant in the presence of vanillin, vanillic acid, protocatechuic acid or a mixture of two or more of them. L'invention concerne un procédé pour produire industriellement une forme unique de l'acide 3-carboxy-cis,cis-muconique et/ou de la 3-carboxymuconolactone à partir d'un mélange de faible poids moléculaire obtenu à partir d'un constituant végétal et contenant de la vanilline, de l'acide vanillique, de l'acide protocatechuique et des analogues grâce à une réaction enzymatique en plusieurs étapes. L'invention concerne en particulier : un plasmide recombinant portant un gène de vanillate déméthylase (gène vanAB), un gène de benzaldéhyde déshydrogénase (gène ligV) et un gène de protocatechuate 3,4-dioxygénase (gène pcaHG); un transformé ayant le plasmide introduit à l'intérieur ; et un procédé de production de l'acide 3-carboxy-cis,cis-muconique et/ou de la 3-carboxymuconolactone, comprenant l'étape de mise en culture du transformé en présence de vanilline, d'acide vanillique, d'acide protocatechuique ou d'un mélange de deux ou plus de ceux-ci.
