Disclosed is a process for separation and purification of histidine from a culture containing the amino acid. According to the process, histidine can be separated and purified by applying a culture containing histidine and a microorganism cell onto the top part of a column packed with a particulate carrier having a particle diameter of 350 µm or more and capable of adsorbing histidine thereon, and then applying an eluent to the column. In the process, it is preferred to use a strongly acidic cation exchange resin as the particulate carrier. L'invention concerne un procédé de séparation et de purification de l'histidine contenue dans un milieu de culture contenant cet acide aminé. Selon ce procédé, l'histidine peut être séparée et purifiée par application d'un milieu de culture, contenant l'histidine et une cellule de micro-organisme, sur la partie supérieure d'une colonne remplie d'un support particulaire, qui présente un diamètre de particules supérieur ou égal à 350 µm et est capable d'adsorber l'histidine appliquée dessus, puis par introduction d'un éluant dans la colonne. Selon ledit procédé, il est préférable d'utiliser une résine échangeuse de cations fortement acide comme support particulaire.
