Disclosed are a method for producing a hybridoma capable of producing a monoclonal antibody directed against a cell membrane surface antigen; a method for producing a monoclonal antibody directed against a cell membrane surface antigen by using the hybridoma; a method for determining the affinity of an antibody capable of binding to a cell membrane surface antigen for the antigen; and a method for assaying or screening an antibody capable of binding to a cell membrane surface antigen by utilizing the determination method. In the immunization of an animal for the production of an antibody, the immunization with, as a sensitizing antigen, a cell strain derived from an animal belonging to the different order from that of the animal to be immunized which can express a cell membrane surface antigen and the immunization with, as a sensitizing antigen, a cell strain derived from an animal belonging to the same order as that of the animal to be immunized in which the cell membrane surface antigen is expressed on the surface of the cell membrane by genetic recombination are combined with each other. In the determination of the affinity of the antibody for the antigen, floating cells presenting the antigen on the surface of the cell membrane thereof are used and the B/F separation of the cells is made by centrifugation or on a filter through which the cells cannot pass. L'invention concerne un procédé de production d'un hybridome capable de produire un anticorps monoclonal dirigé contre un antigène de surface de la membrane cellulaire ; un procédé permettant de produire un anticorps monoclonal dirigé contre un antigène de surface de la membrane cellulaire à l'aide de l'hybridome ; un procédé permettant de déterminer l'affinité d'un anticorps capable de se lier à un antigène de surface de la membrane cellulaire pour l'antigène; et un procédé permettant de doser ou de cribler un anticorps capable de se lier à un antigène de surface de la membrane cellulaire au moyen du procédé de détermination.
