Summary:For RNAi, use is made of a double-stranded RNA for interference which comprises an antisense RNA complementary to one region in the mRNA of a target gene and a sense RNA homologous with the above region in the mRNA of the above target gene, characterized in that the antisense RNA and the sense RNA as described above together form a double-strand over consecutive 25 bases, 26 bases, 27 bases or 28 or more bases. Also, use can be made of a double-stranded RNA having base substitution or base insertion. By using this double-stranded RNA for interference as described above, RNAi with relieved cytotoxicity can be induced at a high efficiency. L'invention concerne l'ARNi avec l'utilisation d'un ARN bicaténaire interférant comportant un ARN anti-sens complémentaire d'une région de l'ARNm d'un gène cible et un ARN sens homologue de ladite région de l'ARNm dudit gène cible. Selon l'invention, ledit ARN anti-sens et ledit ARN sens forment ensemble un double brin présentant consécutivement 25 bases, 26 bases, 27 bases 28 bases ou davantage de bases. De plus, un ARN bicaténaire présentant une substitution de bases ou une insertion de bases peut être utilisé. On peut ainsi induire l'ARNi à toxicité réduite de manière efficace, en utilisant ledit ARN bicaténaire interférant.
