Summary:It is intended to provide cell−free protein synthesis means in a wheat germ system with the use of a base sequence sustaining a translation activity−promoting function originating in CrPV−like viruses. More specifically speaking, a reading frame of a gene is ligated to the downstream of a specific higher−order structure of CrPV−like viruses so that translation of a protein can be initiated from various amino acids in the wheat germ system. Further, a base sequence of about 30 bp or more is introduced between the higher−order structure and a target gene. Thus, overexpression can be established in the cell−free protein synthesis means with the use of wheat germ. L'invention vise à créer un dispositif de synthèse de protéines sans cellules dans un système de germe de blé utilisant une séquence de base qui prend en charge une fonction de promotion de l'activité de traduction qui commence dans des virus similaires à CrPV. Elle concerne plus particulièrement une trame de lecture d'un gène qui est liée à une partie aval d'une structure spécifique d'ordre supérieur des virus similaires à CrPV, de façon que la traduction d'une protéine puisse commencer depuis des acides aminés différents dans le système de germe de blé. En outre, une séquence de base de 30 bp ou plus est introduite entre la structure d'ordre supérieur et un gène cible. De cette manière, on peut assurer la surexpression dans le dispositif de synthèse de protéines sans cellules, qui utilise un germe de blé.
