Parenteral compositions of oil-coated-Amphotericin B in structured-emulsion form, having LD50 of atleast 400mg/kg in mice and process for making the same have been described. The process essentially requires dispersing Amphotericin B in oily vehicle and dispersing emulsifier in the aqueous phase. The process of present invention is simple, cost effective and gives a stable product suitable for parenteral administration. Amphotericin B emulsion compositions prepared by the process of the present invention may be administered to human beings and animals for the treatment of fungal infections, with substantially equivalent or greater efficacy and low drug toxicity as compared to the conventional composition containing Amphotericin B and sodium desoxycholate. Cette invention, qui a trait à des compositions à administrer par voie parentérale d'amphotéricine B enrobée d'huile se présentant sous forme d'émulsion structurée dont la dose létale 50 % est de 400 mg/kg chez la souris, concerne également un procédé de production de ces compositions. Ce procédé consiste à disperser l'amphotéricine B dans un excipient huileux et à disperser un agent émulsifiant dans la phase aqueuse. Ce procédé, qui est simple à mettre en oeuvre et efficace en termes de coût, permet d'obtenir un produit se prêtant bien à une administration parentérale. Il est possible d'administrer ces compositions à base d'émulsion d'amphotéricine B à des êtres humains ainsi qu'à des animaux atteints d'infections fongiques et ce, avec une efficacité sensiblement équivalente ou supérieure ainsi qu'une toxicité inférieure en comparaison de celles des compositions classiques renfermant de l'amphotéricine B et du désoxycholate de sodium.
