Summary:A method is provided for the rapid, substantially isostatic, segregation and amplification of the sequence information of a target nucleic acid sequence positioned within a single- or double-stranded polynucleotide. The method is based on the serial generation of double-stranded DNA engineered to contain terminal nicking sites, nicking of those sites, and extensions from those nicks, thereby displacing any existing polynucleotides. Further provided is a method for detecting polynucleotides using the method of the invention. A kit combining the components commonly used in practicing the method of the invention is also provided.
