Summary:According to some aspects, systems and methods for characterizing phosphoserine-containing polypeptides are described. In some embodiments, a phosphoserine residue of a polypeptide may be converted to a dehydroalanine residue. Some aspects are directed to a method of reacting a polypeptide comprising at least one dehydroalanine residue with a protected thiol compound comprising a click chemistry handle (e.g., an azide, an alkyne) to provide a functionalized polypeptide comprising a click chemistry handle. Some aspects are directed to systems and methods of characterizing phosphoserine residues in polypeptides (e.g., identifying and/or determining the location of a phosphoserine residue) and/or methods of preparing samples for such systems and methods.
