The present invention discloses a biochemical sensing specimen with surface modification and its manufacturing method. The biochemical sensing specimen with surface modification includes an insulating substrate, an electrode system, an insulating pad layer, a reacting layer, and an upper lid. According to the functional definition, the insulating substrate further includes a connecting area and a sensing area. The insulating substrate has one surface coated with the electrode system. The insulating pad layer covers a part of the insulating substrate, wherein the connecting area and the reacting area are formed on the part of the insulating substrate that is not covered by the insulating pad layer. The biochemical sensing specimen with surface modification is characterized in that the electrode system surface of the reacting area is performed with a modification processing to achieve a charged surface with uniform and hydrophilic function.
