An antibiotic effective against gram positive-, gram negative-, pathogenic- and acid fast bacteria, the crystalline free base SF-733 substance being soluble in water, slightly soluble in methanol, sparingly soluble in ethanol, substantially insoluble in n-propanol, n-butanol, acetone, ethyl acetate, benzene, petroleum ether and chloroform, exhibiting in aqueous solution no absorption maximum of ultraviolet spectrum except end absorption, exhibiting infra-red absorption peculiar to amino-glycosidic antibiotics as shown in Fig. 5, in the form of white needles or short rods, fusing while decomposing with effervescence at 192-195 C., giving a positive reaction to Molisch, anthione ninhydrin and #-naphthol-sulphuric acid and negative Fehling, ferric chloride, Brady, biuret, Sakaguchi and maltol, containing C 44À19%, H 7.39%, N 11-60%, 0 (by difference) 36À82%, molecular formula C 17 H 34 N 4 O 10 , chemical structure of 0-#-D-ribofuranosyl 1-(1#5)-O- [α-2,6 -diamino - 2,6 - dideoxy - D - glucopyranosyl- (1#4)].2-deoxystreptamine which is dextro-rotatory and exhibits [α]23D + 42 degrees (C = 1, H 2 0) is prepared by cultivating Streptomyces ribosidificus SF-733 (ATCC 21294) in an aqueous nutrient medium containing assimilable nitrogen and carbon sources under submerged aerobic conditions to accumulate the amorphous free base of SF-733 substance in the medium and then recovering the SF-733 substance. Amorphous SF-733 substance may be recovered from the culture medium by filtration followed by column chromatography and crystalline SF-733 is produced by employing alcoholic solvents. The sulphate salt of SF-733 substance is formed by reaction of the free base with sulphuric acid. |
