Summary:FIELD: medicine, medicinal microbiology. SUBSTANCE: the present innovation deals with obtaining cell-free pertussis vaccine. In liquid nutritive medium one should cultivate pertussis bacteria, separate supernatant to remove toxic material, detoxication of supernatant should be carried out due to successive addition of saccharose up to 3-5% concentration and formalin up to 0.4% concentration followed by keeping the mixture at 36 C for 3 d. Partially neutralized toxic material is removed out of supernatant at the presence of 1%-sodium hexametaphosphate solution and 2n trichloroacetic acid at pH being 3.4-3.5. The residue obtained should be separated due to centrifuging at 12500 rot./min for 60 min to be solved in 0.07 M phosphate buffer at pH being 7.5. The preparation obtained should be supplemented with saccharose up to 10% concentration and formalin up to 0.15-0.2% concentration. It should be finally detoxicated at 36 C for 15 d. As a result, a highly active and low toxic Bordetella pertussis anatoxin is obtained to be applied for producing cell-free pertussis vaccine. EFFECT: higher efficiency. 1 cl, 2 ex
