Summary: | biotechnology. SUBSTANCE: method involves preparing inoculum material and industrial culturing the strain Brucella abortus 19 in liquid nutrient medium containing Hottinger's broth, glycerol, glucose, NaCl, yeast autolysate and water. Culturing is carried out under submerged conditions for 19-21 h at regulation of partial pressure of oxygen dissolved in cultural fluid for the all culturing process. After addition of inoculum material the levels of 20-25 pO2, 40-45 pO2 and 30-35 pO2 are adjusted from 1 to 6 h, from 6 to 16-18 h and then from 16-18 to 19-21 h, respectively. Grown bacterial cells are separated and concentrated by ultrafiltration on fibers with retention limit 15-50 kDa, inactivated by heating, stained with rose Bengal dye, resuspended in buffer diluting agent and the end product is standardized. Invention can be used for industrial production of preparations for diagnosis of brucellosis. EFFECT: simplified technology, decreased time for preparing color brucellosis antigen for rose Bengal specimen, improved quality. 3 ex |
