Summary: | FIELD: molecular biology, genetic engineering, medicine. SUBSTANCE: invention relates to the constructed recombinant plasmid DNA that can be used in biotechnology for preparing human insulin. Recombinant DNA based on plasmid pKK 223-3 and encoding preproinsulin is inserted between restriction sites Eco RI and Hind III. Triplet encoding methionine is replaced for triplet that encodes arginine. This ensures to obtain proinsulin by enzymatic cleavage.
